T-3861174 is a novel small molecule prolyl tRNA synthetase (PRS) with binding IC50 of 2.3 nM; induces cell death in several tumor cell lines including SK-MEL-2 (IC50=0.1 uM) without complete suppression of translation, T-3861174-induced cell death was caused by activation of the GCN2-ATF4 pathway; exhibits significant tumor growth inhibition inSK-MEL-2, G-361, and HT-1080 xenograft models without severe body weight losses.
