A specific stain for alpha-glucosidases in isoelectric focusing gels.

Abstract

α-Glucosidases separated on isoelectric focusing gels can be localized by a two-step staining procedure. First, bromonaphthyl-α- d-glucoside is cleaved by the α-glucosidases in the gel; then, the released bromonaphthol is precipitated with Fast Blue B salt. The procedure is specific for α-glucosidases, gives sharp enzyme bands, and does not inactivate the α-glucosidases. Using this procedure, it has been demonstrated that the maltase and α-methylglucosidase induced by maltose are separate enzymes.
