Abstract

The tautomerase superfamily consists of homologous proteins with a characteristic βαβ structural fold and a conserved catalytic N-terminal proline.[1] 4-Oxalocrotonate tautomerase (4-OT) from Pseudomonas putida mt-2 is a member of this superfamily, and catalyses the enol-keto tautomerisation of 2-hydroxyhexa-2, 4-dienedioate(1) to 2-oxohex-3-enedioate(2; Scheme 1).[2] In this natural activity of 4-OT, the Pro1 residue functions as the catalytic ...
